Tissue Microarrays

The CPCTR has slides from six prostate cancer TMAs with associated clinical data.

TMAs Available:

  • TMA 1: Archival TMA slide set based on longest follow up data available from the resource
  • TMA 2: Gleason TMA slide set based on Gleason Grade distribution
  • TMA 3: Hormone Refractory TMA slide set
  • TMA 4: Ethnicity TMA slide set: 150 African American and 150 Caucasian cases
  • TMA 5: Outcome TMA
  • TMA 6: Metastatic TMA

TMA 1

This TMA contains the 299 cases with the longest potential follow-up time (year of prostatectomy 1990-1992).

  • Cancer tissue from radical prostatectomy specimens of 299 patients
  • Control non-neoplastic tissue from benign prostatic hyperplasia (BPH)
  • Control non-diseased tissue from organ donor prostates
  • Cores from two prostate cancer cell lines: LNCaP and PC-3

TMA 2 (Gleason TMA)

This TMA was designed to investigate differences in the prevalence of a potential marker among different Gleason grades. The arrays were designed by National Cancer Institute statisticians to provide high statistical power for studies of grade-specific markers of prostate cancer.

  • Constructed by using breakdown by Gleason score:
    • Gleason 4: 3 Cases
    • Gleason 5: 31
    • Gleason 6: 53
    • Gleason 7: 54
    • Gleason 8: 52
    • Gleason 9: 54
    • Gleason 10: 2
  • Cancer tissue from radical prostatectomy specimens of 250 patients, of which 58 are also represented with HGPIN cores.
  • Control non-neoplastic tissue from 18 benign prostatic hyperplasia (BPH) cases.
  • Control non-diseased tissue from 14 organ donor prostates.
  • Cores from three prostate cancer cell lines: LNCaP, DU-145, and PC-3.

TMA 3 (Hormone Refractory TMA)

This TMA is designed to investigate differences in the prevalence of a potential marker between hormone refractory and hormone naive prostate cancer. Tissue is assigned as hormone refractory if the surgery took place at least 6 months after orchiectomy or initiation of medical hormone therapy. The majority of refractory cases had two consecutive rising PSA values after hormone therapy or a distant metastatic site identified after therapy.

  • Constructed by matching hormone refractory tissues with hormone naive tissues using patient age, race, and Gleason grade:
    • Hormone Refractory Prostate Tissue: 29 cases
    • Hormone Naive Prostate Tissue: 29 cases
    • Hormone Refractory Metastatic Tissue: 14 cases
    • Hormone Naive Metastatic Tissue: 14 cases
  • Prostate cancer tissue from transurethral resection of the prostate (TURP) and radical prostatectomy.
  • Metastatic cancer tissue from multiple distant sites including bone, regional lymph nodes, and bladder.
  • Control non-neoplastic tissue from 6 benign prostatic hyperplasia (BPH) cases.
  • Control non-diseased tissue from 14 organ donor prostates.
  • Control non-diseased tissue from 4 testes.
  • Cores from three prostate cancer cell lines: LNCaP, DU-145, and PC-3.

TMA 4 (Ethnicity TMA)

This TMA is designed to investigate possible differences in marker prevalence between African Americans and Caucasian Americans with prostrate cancer. The TMA includes four neoplastic tissue samples from each of 150 African American prostate cancer cases, with an equal number of neoplastic samples from 150 Caucasian cases. In addition, the Ethnicity TMA (TMA4) contains 17 BPH cores, 13 normal cores, and 3 cell line cores (LnCAP, DU-145, PC3) on each slide of the 4-slide set.

TMA 5 (Outcome TMA)

This TMA is designed to identify markers associated with prostate cancer recurrence after prostatectomy. Approximately 202 paired recurrence and non-recurrence cases (404 altogether), with individual pairs matched based on race, Gleason sum score (including 3+4 vs. 4+3), pT stage, pN stage, pM stage, and age at prostatectomy.

TMA 6 (Metastatic TMA)

This TMA is designed to identify markers differentially expressed in primary prostate cancer lesions versus metastatic lesions. For 21 patients, tissue cores from both the primary lesion and the metastatic lymph node lesion are included. For 20 patients, tissue cores from metastatic lesion only, and for 12 patients, tissue cores from the primary lesion only (from patients who also had lymph node metastases at time of resection), are included, as well as cell line and normal donor controls.

The information provided for each case on the array includes over 20 data elements including: age at diagnosis, race, PSA at diagnosis, tumor size, TNM stage, Gleason score and grade, recurrence, and vital status.

All researchers may inquire about TMA slides (academic or commercial, US or foreign).

Cost for Tissue Microarray (TMA) Slides:

  • $1800 for a set of four slides, with annotation (includes 3 free optimization TMA slides for testing probe)
  • $300 for each additional TMA slide, with annotation
  • $75 per additional optimization TMA slide
